Recently, linear regression analysis (LRA) of constitutive activity as a function of TSHR expression determined by 125I-bTSH binding or FACS analysis compared to wt TSHR was described as a more reliable way of characterizing the in vitro activity (IVA) of constitutively activating TSHR mutations. Therefore, we analyzed the TSHR mutations published LRAs by a systematic review of the literature. The published LRAs of 10 different sporadic TSHR germline mutations, of 10 different familial TSHR germline mutations, of 39 different somatic TSHR mutations described in 170 hot nodules and the published LRAs of 3 different somatic TSHR mutations reported for 3 hot thyroid carcinomas were compared by non-parametric tests. The four groups showed significantly different LRAs (mean ranks: sporadic: 133,21; familiar: 83,02; TTN: 124,90; hot carcinomata: 134,17; Kruskal-Wallis-Test: asymp. Sig.: 0,002). Furthermore, in pairwise testings sporadic mutations had a significantly higher LRA than familiar mutations (mean ranks: sporadic: 38,29; familiar: 27,88; Mann-Whitney-Test: asymp. Sig, 2-tailed: 0,054) and somatic TSHR mutations of TTNs showed higher activity than familiar mutations (mean ranks somatic: 118,07; familiar: 78,33; asymp. Sig. 2-tailed&lt;0,0001). For the 3 different TSHR mutations of hot follicular carcinomas non-parametric testing showed no significant difference to the LRA of the sporadic, familiar or somatic mutations in TTNs (asymp. Sig (2-tailed): 0,884; 0,165; 0,815 respectively.) This systematic review of published LRAs provides evidence for a higher IVA of sporadic as opposed to familiar TSHR germline mutations thus supporting the analogous clinical impression.
