The same substrates (Fe2+ and H2O2) may be used to experimentally induce oxidative damage to macromolecules. External H2O2 has recently been found to be not indispensable for experimental induction of lipid peroxidation in porcine ovary homogenates. The aim of the study was to evaluate the effects of Fe2+ and\/or H2O2 on lipid peroxidation in porcine thyroid homogenates. Thyroid homogenates were incubated in the presence of either H2O2 (100, 50, 10, 5.0, 1.0, 0.5, 0.1, 0.01, 0.001, 0.0001, and 0.00001 mM) or FeSO4 (300, 150, 75, 30, 15, 7.5, 3.0, 1.5, and 0.75 ?M), or in the presence of those two factors used together, namely, in the presence of FeSO4 (30 ?M) plus H2O2 (100, 50, 10, 5.0, 1.0, 0.5, 0.1, 0.01, 0.001, 0.0001, and 0.00001 mM), or in the presence of H2O2 (0.5 mM) plus FeSO4 (300, 150, 75, 30, 15, 7.5, 3.0, 1.5, and 0.75 ?M). The level of lipid peroxidation was measured spectrophotometrically and expressed as the amount of MDA + 4-HDA (nmol) per mg of protein. Either H2O2 alone or Fe2+ alone did not affect lipid peroxidation in porcine ovary homogenates at all. When Fe2+ and H2O2 were applied together, the level of lipid peroxidation increased significantly, however, that effect did not depend of H2O2 concentration at all, but it did clearly depend on Fe2+ concentration, with statistical significance for the highest used concentrations (300, 150, 75, 30, and 15 ?M).
