Therefore the aim of this project was to determine the expression of different members of this pathway and to deep inside on the regulation of beta-catenin (bcat) in differentiated and tumour thyroid cells. RNA was extracted from differentiated rat thyroid cells (PCCl3), treated with TSH and IGF1 for different periods of time or from human papilar (TPC1), poorly differentiated (NPA), follicular (WRO), and anaplastic (FRO and ARO) carcinoma tumour cells. Gene expression was analyzed by QRT-PCR and total, nuclear or cytosolic proteins by Western blot. Subcellular localization of bcat was confirmed by confocal microscopy. Transcriptional activation was determined either from bcat, using its dependent promoter TopFlash, or from thyroid specific gene NIS, TPO and Tg promoters. We have observed differential expression of the Wnt pathways members between differentiated and tumours thyroid cells. Interestingly, Wnt7A and Wnt10B, two members of the canonical pathway, were expressed exclusively in tumour cells and in these cells bcat is aberrantly located outside of the plasma membrane. This fact correlates with a higher bcat transcriptional activity in the most undifferentiated tumour cells. In addition we show that TSH, a hormone that regulates thyroid cells proliferation induces nuclear bcat translocation as well as its transcriptional activity. Regulation of thyroid differentiation genes by bcat was also found. We can conclude that there is a correlation between bcat activity and the degree of thyroid cell dedifferentiation implying the canonical Wnt\/bcat pathway in thyroid transformation.<\/p><\/div>\n","protected":false},"excerpt":{"rendered":"<p>Sastre-Perona A., Moya C.
