Its action is initiated by the activation of T4 to T3, catalyzed by the type 1 or the type 2 deiodinases (D1 or D2). Irreversible inactivation of T4 and T3 is catalyzed by the type 3 deiodinase D3, the major physiological thyroid hormone inactivator. Skin is a major target for thyroid hormone and local intracellular concentration is modulated by the balance between activating (D2) and inactivating (D3) deiodinases. In skin, constitutive Shh signaling causes basal cell carcinoma (BCCs) the most common cancer in humans. We have recently demonstrated that Shh-Gli2 pathway induces D3 in human and mouse BCC, however the role of D3 in BCC tumour formation and maintenance is unknown. In the present study we used a mouse BCC cell line (G2N2c) as working model to investigate the role of local control of TH action- in the Shh-driven tumorigenesis. G2N2c were grown in media deprived of thyroid hormone (CS), or CS plus exogenous T3 at various concentrations. We observed a reduction in the growing rate inversely correlated to the T3 in the media. Furthermore, T3 interferes specifically and directly with Gli2 expression, by reducing Gli2 protein level and activity. Finally, to study the effect of thyroid hormone on the BCC tumour growth in vivo, we injected nude mice with BCC cells and treated the resulting tumors with a T3-based cream. Tumour weight, measured 15 days after the injection, showed a significative reduction in tumor topically T3-treated versus controls. In conclusion, this work sheds light on a functional cross-talk between thyroid hormone and Shh-signalling in BCC. This mechanism could have broad implications, and may lead to the identification of novel therapeutic approaches aimed at interfering with these signals in a pathological context.

Dentice M.1, Luongo C.1, Ambrosio R.1, Aristarco V.1, Casillo A.1, D?Ambrosio R.1, Mansi C.1, Fenzi G.1, Larsen PR.2, Salvatore D.1 1Dipartimento di Endocrinologia ed Oncologia Molecolare e Clinica, Universit? di Napoli “Federico II”, Naples, Italy, 2Division of Endocrinology, Brigham and Womens Hospital, Harvard, Boston, USA Abstract Thyroid hormone is a critical mediator of cellular metabolism Read More

[45,2,9

post-1285","post","type-post","status-publish","format-standard","hentry","category-volume-7-issue-3","category-journal-articles","category-volume-7
["GET
